NM_022051.3(EGLN1):c.331A>G (p.Lys111Glu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K111E variant (also known as c.331A>G), located in coding exon 1 of the EGLN1 gene, results from an A to G substitution at nucleotide position 331. The lysine at codon 111 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
