NM_001430.5(EPAS1):c.1004A>G (p.Gln335Arg) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q335R variant (also known as c.1004A>G), located in coding exon 8 of the EPAS1 gene, results from an A to G substitution at nucleotide position 1004. The glutamine at codon 335 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
