NM_000548.5(TSC2):c.1805A>C (p.Tyr602Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y602S variant (also known as c.1805A>C), located in coding exon 16 of the TSC2 gene, results from an A to C substitution at nucleotide position 1805. The tyrosine at codon 602 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
