NM_020975.6(RET):c.1334C>G (p.Ser445Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S445C variant (also known as c.1334C>G), located in coding exon 7 of the RET gene, results from a C to G substitution at nucleotide position 1334. The serine at codon 445 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
