Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005585.5(SMAD6):c.766G>A (p.Gly256Ser), citing Ambry Variant Classification Scheme 2023: The p.G256S variant (also known as c.766G>A), located in coding exon 1 of the SMAD6 gene, results from a G to A substitution at nucleotide position 766. The glycine at codon 256 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
