Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.4225T>G (p.Ser1409Ala), citing Ambry Variant Classification Scheme 2023: The p.S1409A variant (also known as c.4225T>G), located in coding exon 18 of the NPAT gene, results from a T to G substitution at nucleotide position 4225. The serine at codon 1409 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
