NM_000209.4(PDX1):c.190G>T (p.Asp64Tyr) was classified as Uncertain significance for Maturity-onset diabetes of the young by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D64Y variant (also known as c.190G>T), located in coding exon 1 of the PDX1 gene, results from a G to T substitution at nucleotide position 190. The aspartic acid at codon 64 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
