NM_000492.4(CFTR):c.4093A>G (p.Lys1365Glu) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1365E variant (also known as c.4093A>G), located in coding exon 25 of the CFTR gene, results from an A to G substitution at nucleotide position 4093. The lysine at codon 1365 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
