NM_000492.4(CFTR):c.425T>G (p.Ile142Ser) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I142S variant (also known as c.425T>G), located in coding exon 4 of the CFTR gene, results from a T to G substitution at nucleotide position 425. The isoleucine at codon 142 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
