NM_000245.4(MET):c.3408T>A (p.His1136Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1154Q variant (also known as c.3462T>A), located in coding exon 16 of the MET gene, results from a T to A substitution at nucleotide position 3462. The histidine at codon 1154 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
