NM_006904.7(PRKDC):c.1964T>G (p.Leu655Trp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L655W variant (also known as c.1964T>G), located in coding exon 18 of the PRKDC gene, results from a T to G substitution at nucleotide position 1964. The leucine at codon 655 is replaced by tryptophan, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
