NM_004360.5(CDH1):c.137T>G (p.Leu46Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L46R variant (also known as c.137T>G), located in coding exon 2 of the CDH1 gene, results from a T to G substitution at nucleotide position 137. The leucine at codon 46 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
