NM_000179.3(MSH6):c.217A>C (p.Asn73His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N73H variant (also known as c.217A>C), located in coding exon 1 of the MSH6 gene, results from an A to C substitution at nucleotide position 217. The asparagine at codon 73 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
