NM_053025.4(MYLK):c.3868G>C (p.Glu1290Gln) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1290Q variant (also known as c.3868G>C), located in coding exon 20 of the MYLK gene, results from a G to C substitution at nucleotide position 3868. The glutamic acid at codon 1290 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
