Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.359G>T (p.Cys120Phe), citing Ambry Variant Classification Scheme 2023: The p.C120F variant (also known as c.359G>T), located in coding exon 2 of the BLM gene, results from a G to T substitution at nucleotide position 359. The cysteine at codon 120 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
