NM_001082486.2(ACD):c.108G>C (p.Gln36His) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q122H variant (also known as c.366G>C), located in coding exon 2 of the ACD gene, results from a G to C substitution at nucleotide position 366. The glutamine at codon 122 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
