Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.150G>T (p.Lys50Asn), citing Ambry Variant Classification Scheme 2023: The p.K50N variant (also known as c.150G>T), located in coding exon 2 of the NF1 gene, results from a G to T substitution at nucleotide position 150. The lysine at codon 50 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
