Pathogenic for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.5516dup (p.Gln1841fs), citing Ambry Variant Classification Scheme 2023: The c.5453dupT pathogenic mutation, located in coding exon 37 of the NF1 gene, results from a duplication of T at nucleotide position 5453, causing a translational frameshift with a predicted alternate stop codon (p.Q1820Pfs*21). This alteration has been observed in at least one individual with a diagnosis of neurofibromatosis type 1 and a GIST (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
