Submitter rationale: The c.1226dupA pathogenic mutation, located in coding exon 12 of the MLH1 gene, results from a duplication of A at nucleotide position 1226, causing a translational frameshift with a predicted alternate stop codon (p.A410Gfs*7). This variant has been identified in a proband whose Lynch syndrome-associated tumor demonstrated loss of MLH1/PMS2 expression by immunohistochemistry (Ambry internal data).
