This variant lies in the FLCN gene (transcript NM_144997.7) at 4 bases into the intron immediately after coding-DNA position 1176, deleting one base. Submitter rationale: The c.1176+4delA intronic variant, located in intron 7 of the FLCN gene, results from a deletion of one nucleotide within intron 7 of the FLCN gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and will result in the creation or strengthening of a novel splice donor site. RNA studies have demonstrated that this alteration results in an incomplete splice defect; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
