Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.509A>C (p.His170Pro), citing Ambry Variant Classification Scheme 2023: The p.H170P variant (also known as c.509A>C), located in coding exon 5 of the PMS2 gene, results from an A to C substitution at nucleotide position 509. The histidine at codon 170 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
