Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.633A>C (p.Lys211Asn), citing Ambry Variant Classification Scheme 2023: The p.K211N variant (also known as c.633A>C), located in coding exon 5 of the RAD50 gene, results from an A to C substitution at nucleotide position 633. The lysine at codon 211 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
