Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_017950.4(CCDC40):c.1270A>G (p.Lys424Glu), citing Ambry Variant Classification Scheme 2023: The p.K424E variant (also known as c.1270A>G), located in coding exon 8 of the CCDC40 gene, results from an A to G substitution at nucleotide position 1270. The lysine at codon 424 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
