Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.3321A>T (p.Glu1107Asp), citing Ambry Variant Classification Scheme 2023: The p.E1107D variant (also known as c.3321A>T), located in coding exon 9 of the BRCA1 gene, results from an A to T substitution at nucleotide position 3321. The glutamic acid at codon 1107 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
