NM_001148.6(ANK2):c.3748G>A (p.Gly1250Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1250S variant (also known as c.3748G>A), located in coding exon 31 of the ANK2 gene, results from a G to A substitution at nucleotide position 3748. The glycine at codon 1250 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
