Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005120.3(MED12):c.2643A>C (p.Glu881Asp), citing Ambry Variant Classification Scheme 2023: The p.E881D variant (also known as c.2643A>C), located in coding exon 19 of the MED12 gene, results from an A to C substitution at nucleotide position 2643. The glutamic acid at codon 881 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
