Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_015450.3(POT1):c.723C>G (p.Ile241Met), citing Ambry Variant Classification Scheme 2023: The p.I241M variant (also known as c.723C>G), located in coding exon 6 of the POT1 gene, results from a C to G substitution at nucleotide position 723. The isoleucine at codon 241 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
