Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_030777.4(SLC2A10):c.848C>G (p.Ala283Gly), citing Ambry Variant Classification Scheme 2023: The p.A283G variant (also known as c.848C>G), located in coding exon 2 of the SLC2A10 gene, results from a C to G substitution at nucleotide position 848. The alanine at codon 283 is replaced by glycine, an amino acid with similar properties. This variant co-occurred with a variant in the COL3A1 gene in an individual from a thoracic aortic aneurysm and dissection cohort; however, details were limited (Kathiravel U et al. Mol. Cell. Probes, 2013 Apr;27:103-8). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
