NM_004006.3(DMD):c.8308G>C (p.Asp2770His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D2770H variant (also known as c.8308G>C), located in coding exon 56 of the DMD gene, results from a G to C substitution at nucleotide position 8308. The aspartic acid at codon 2770 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
