Uncertain significance — the classification assigned by Ambry Genetics to NM_013266.4(CTNNA3):c.367C>G (p.Gln123Glu), citing Ambry Variant Classification Scheme 2023: The p.Q123E variant (also known as c.367C>G), located in coding exon 3 of the CTNNA3 gene, results from a C to G substitution at nucleotide position 367. The glutamine at codon 123 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
