Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.10303C>T (p.His3435Tyr), citing Ambry Variant Classification Scheme 2023: The p.H3006Y variant (also known as c.9016C>T), located in coding exon 34 of the OBSCN gene, results from a C to T substitution at nucleotide position 9016. The histidine at codon 3006 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
