NM_005751.5(AKAP9):c.10500A>C (p.Glu3500Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E3500D variant (also known as c.10500A>C), located in coding exon 42 of the AKAP9 gene, results from an A to C substitution at nucleotide position 10500. The glutamic acid at codon 3500 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
