Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.7219G>C (p.Glu2407Gln), citing Ambry Variant Classification Scheme 2023: The p.E2407Q variant (also known as c.7219G>C), located in coding exon 31 of the AKAP9 gene, results from a G to C substitution at nucleotide position 7219. The glutamic acid at codon 2407 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
