NM_000264.5(PTCH1):c.4299G>C (p.Gln1433His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1433H variant (also known as c.4299G>C), located in coding exon 23 of the PTCH1 gene, results from a G to C substitution at nucleotide position 4299. The glutamine at codon 1433 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
