NM_000264.5(PTCH1):c.3824G>C (p.Arg1275Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1275T variant (also known as c.3824G>C), located in coding exon 23 of the PTCH1 gene, results from a G to C substitution at nucleotide position 3824. The arginine at codon 1275 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
