This variant lies in the FLNA gene (transcript NM_001110556.2) at coding-DNA position 827, where G is replaced by A; at the protein level this means replaces arginine at residue 276 with glutamine — a missense variant. Submitter rationale: The p.R276Q variant (also known as c.827G>A), located in coding exon 4 of the FLNA gene, results from a G to A substitution at nucleotide position 827. The arginine at codon 276 is replaced by glutamine, an amino acid with highly similar properties. Based on data from gnomAD, the A allele has an overall frequency of 0.0006% (1/181615) total alleles studied, with no hemizygotes observed. The highest observed frequency was 0.0012% (1/81070) of European (non-Finnish) alleles. This amino acid position is well conserved in available vertebrate species.
