Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000393.5(COL5A2):c.3737A>G (p.Asp1246Gly), citing Ambry Variant Classification Scheme 2023: The p.D1246G variant (also known as c.3737A>G), located in coding exon 51 of the COL5A2 gene, results from an A to G substitution at nucleotide position 3737. The aspartic acid at codon 1246 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
