NM_052947.4(ALPK2):c.3312T>A (p.Asp1104Glu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1104E variant (also known as c.3312T>A), located in coding exon 4 of the ALPK2 gene, results from a T to A substitution at nucleotide position 3312. The aspartic acid at codon 1104 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
