NM_017636.4(TRPM4):c.1507G>C (p.Val503Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V503L variant (also known as c.1507G>C), located in coding exon 11 of the TRPM4 gene, results from a G to C substitution at nucleotide position 1507. The valine at codon 503 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
