Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.8545C>G (p.Gln2849Glu), citing Ambry Variant Classification Scheme 2023: The p.Q2849E variant (also known as c.8545C>G), located in coding exon 65 of the FBN2 gene, results from a C to G substitution at nucleotide position 8545. The glutamine at codon 2849 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
