Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 251302 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.152G>T has been observed in at least one individual affected with Arrhythmogenic Right Ventricular Dysplasia/Cardiomyopathy (Headrick_2019). These reports do not provide unequivocal conclusions about association of the variant with Arrhythmogenic Right Ventricular Dysplasia/Cardiomyopathy. One co-occurrence with another pathogenic variant has been reported (DSP c.478C>T, p.R160X*), providing supporting evidence for a benign role (Headrick_2019). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
