Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_181486.4(TBX5):c.1045G>A (p.Glu349Lys), citing Ambry Variant Classification Scheme 2023: The p.E349K variant (also known as c.1045G>A), located in coding exon 8 of the TBX5 gene, results from a G to A substitution at nucleotide position 1045. The glutamic acid at codon 349 is replaced by lysine, an amino acid with similar properties. This alteration has been reported in a dilated cardiomyopathy (DCM) cohort; however, clinical details were limited (Mazzarotto F et al. Circulation, 2020 Feb;141:387-398). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
