This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 137, where G is replaced by A; at the protein level this means replaces glycine at residue 46 with aspartic acid — a missense variant. Submitter rationale: The p.G46D variant (also known as c.137G>A), located in coding exon 2 of the MYBPC3 gene, results from a G to A substitution at nucleotide position 137. The glycine at codon 46 is replaced by aspartic acid, an amino acid with similar properties. This variant was detected in a cardiomyopathy genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
