Uncertain significance — the classification assigned by Ambry Genetics to NM_003628.6(PKP4):c.1373G>C (p.Ser458Thr), citing Ambry Variant Classification Scheme 2023: The p.S458T variant (also known as c.1373G>C), located in coding exon 8 of the PKP4 gene, results from a G to C substitution at nucleotide position 1373. The serine at codon 458 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
