NM_003628.6(PKP4):c.1871G>A (p.Arg624Lys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R624K variant (also known as c.1871G>A), located in coding exon 10 of the PKP4 gene, results from a G to A substitution at nucleotide position 1871. The arginine at codon 624 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
