Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.4975A>G (p.Asn1659Asp), citing Ambry Variant Classification Scheme 2023: The p.N1659D variant (also known as c.4975A>G), located in coding exon 38 of the POLE gene, results from an A to G substitution at nucleotide position 4975. The asparagine at codon 1659 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
