Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_201596.3(CACNB2):c.253G>A (p.Asp85Asn), citing Ambry Variant Classification Scheme 2023: The p.D31N variant (also known as c.91G>A), located in coding exon 2 of the CACNB2 gene, results from a G to A substitution at nucleotide position 91. The aspartic acid at codon 31 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
