Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_020975.6(RET):c.214C>T (p.Leu72Phe), citing Ambry Variant Classification Scheme 2023: The p.L72F variant (also known as c.214C>T), located in coding exon 2 of the RET gene, results from a C to T substitution at nucleotide position 214. The leucine at codon 72 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
