NM_198253.3(TERT):c.610A>T (p.Asn204Tyr) was classified as Uncertain significance for Dyskeratosis congenita by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N204Y variant (also known as c.610A>T), located in coding exon 2 of the TERT gene, results from an A to T substitution at nucleotide position 610. The asparagine at codon 204 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
