Uncertain significance — the classification assigned by Ambry Genetics to NM_018975.4(TERF2IP):c.178G>A (p.Gly60Arg), citing Ambry Variant Classification Scheme 2023: The p.G60R variant (also known as c.178G>A), located in coding exon 1 of the TERF2IP gene, results from a G to A substitution at nucleotide position 178. The glycine at codon 60 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
